Calorimetric & respirometric monitoring of metabolism: some Examples

چکیده
inTroDuCTion Simultaneous measurements of the rates of heat production and oxygen utilisation by living tissues provides a direct quantitative means of detecting subtle changes in metabolic state in the face of altered physiological and environmental conditions. The ratio of calorimetrically measured heat flux to the respirometrically measured oxygen flux is often called the “calorimetric-respirometric ratio” or CR ratio. This can be used to partition total metabolic energy flux into its aerobic and anaerobic components by comparison of the experimental CR ratio with the theoretical “oxycaloric equivalent” for fully aerobic respiration. Such information is most important when evaluating the physiological effect of environmental stress. Industrialisation of many processes has lead to an increasing amount of synthetic and natural waste products expelled into the environment The consequences of this can often be fatal as seen by the many examples of polluted rivers, waste land and the atmosphere. This large amount of waste can be seen to be harmful to the ecosystem and has lead to an imbalance of the natural flora and fauna. There is a need, therefore to study the effects of pollutants and to evaluate quantitatively their toxicological effects. The combined use of microcalorimetry and respirometry provides a useful means of monitoring such effects.

Respirometric oxidative phosphorylation assessment in saponin-permeabilized cardiac fibers.
Investigation of mitochondrial function represents an important parameter of cardiac physiology as mitochondria are involved in energy metabolism, oxidative stress, apoptosis, aging, mitochondrial encephalomyopathies and drug toxicity. Given this, technologies to measure cardiac mitochondrial function are in demand. One technique that employs an integrative approach to measure mitochondrial fun...
متن کاملThe Design and Operation of a MEMS Differential Scanning Nanocalorimeter for High-Speed Heat Capacity Measurements of Ultrathin Films
A MEMS sensor has been developed for use as a calorimetric cell in an ultra-sensitive, thin-film, differential scanning calorimetric technique. The sensor contains a freestanding, thin (30 nm to 1000 nm), low-stress silicon nitride membrane with lateral dimensions of a few millimeters. This membrane, along with a thin (50 nm) metallization layer, forms a calorimetric cell with an exceptionally ...
متن کاملMonitoring of Yeast Metabolism with Calorimetry
A system for calorimetric measurements is established in a 1500 L pilot bioreactor. Saccharomyces cerevisiae, baker’s yeast has three metabolic pathways: 1) purely oxidative, glucose consuming; 2) purely reductive, glucose consuming and ethanol producing and 3) purely oxidative, ethanol consuming. Designing experiments to lead the yeast to a single pathway, the accuracy of the calorimetric meas...
متن کاملThe Use of a Respirometric Technique for Assessing the Evolution of the Stability of a Sewage Sludge Compost Developed under Full-Scale Conditions
Composting is one of the best ways to properly manage the high production of sewage sludge with the aim of obtaining sanitized products with a stable organic matter. The evolution of the stability of a compost consisting of sewage sludge, straw and sawdust, developed under full-scale conditions, was analyzed. The stability was assessed by a respirometric assay performed for 30 days, where the O...

A number of water tunnel respirometer types exist (Cech, 1990; Lucas et al., 1993), but the ...
